Map the comparison

A variable is a property that can take different values. The independent variable is the condition you deliberately vary; the outcome is what you measure; controlled variables are conditions you keep comparable; and a confounder is a second difference that could explain the outcome. A control is a comparison condition that helps interpret what the test condition did. “Control” does not mean a perfect or zero result: its purpose depends on the question. Define the independent experimental unit—the item or trial receiving the condition—and plan independent replication at that level; repeated readings of one unit do not substitute for new units.

A blank contains no intended analyte or signal and travels through relevant handling to reveal contamination or background. A standard or reference material has a known or assigned value and tests whether a measurement process responds as expected. A control may be a comparison treatment, a blank, or a positive control; the labels are not interchangeable. Define what each check is supposed to detect before running it.

  • Negative control: should not show the effect under the method, if the method is working.
  • Positive control: should show a known response and demonstrates that the method can detect it.
  • Method blank: clean matrix carried through preparation and analysis to expose process contamination.
  • Standard: known value or concentration used for calibration or verification, with a stated basis.

Plan the checks

Write the expected result, acceptance rule, and response for each check. A blank failure does not automatically mean every sample is unusable; it triggers investigation of contamination, sensitivity, and sample levels. A standard outside its assigned range may indicate drift, preparation error, or a mismatch between method and material. Do not invent universal limits: use the method, specification, or a justified project rule and record its version.

  • Use the same units and basis in samples, standards, and acceptance rules.
  • Keep a failed control visible and link any repeat or deviation to the original record.
  • Do not subtract a blank silently unless the method explicitly requires it.

Worked example

Check roles in a water comparison

The learner compares volume readings from two vessels and wants to ensure a dirty transfer container did not bias one group.

  3. 03Reference check: a vessel with an assigned capacity, used to detect gross drift; the assigned basis and tolerance are recorded.
  4. 04Response: preserve all readings, inspect handling and equipment, and repeat only under a documented rationale.

    Draw the causal map

    List the condition, outcome, fixed conditions, and plausible confounders. Mark what is actually measured.

    Assign check roles

    For each control, blank, or standard, write what it should show, what failure means, and the source of its acceptance rule.

    Write the response

    Specify whether a failed check pauses the run, starts an investigation, or permits a qualified decision with documented limitation.

Common mistakes

Correct the process, preserve the record.

Calling an untreated comparison a blank.

A blank lacks intended analyte or signal; an untreated sample can still contain background material.

Treating a standard as proof every sample is accurate.

A standard checks a defined part of the process and range; document its traceability, matrix, and limits.

Deleting data after a failed control.

Preserve the raw result, investigate the failure, and state which data are affected and why.

Build a control map

Use a supplied table containing two vessel groups, a clean water blank, and a reference-volume check. Design the map for a question about repeatability and possible contamination.

  2. What result would make you investigate before interpreting samples?

01How is a blank different from a negative control?

Answer: A blank is designed to contain no intended analyte or signal and reveals background or contamination. A negative control is a comparison expected not to show the target effect; it may contain other material.

02Why write an acceptance rule before looking at results?

Answer: It reduces outcome-driven decisions and makes the meaning of a failed check explicit. The rule must still be scientifically justified and revisable through a documented method change.

03A standard is in range but the blank is contaminated. Can you simply proceed?

Answer: Not automatically. The standard does not rule out contamination. Investigate the blank, determine which samples could be affected, preserve records, and follow the method or qualified decision process.
