Writing methods another person can follow

A method is a route, not a slogan

‘Pipette 1 mL carefully’ is not a reproducible method. A useful method names the material and identity rule, equipment type and ID, volume and units, vessel, sequence, relevant conditions, timing, endpoint, quality checks, and what to do when the check fails. It can cite a manufacturer manual or validated procedure for routine details, but it must say which version was followed and describe any deviation. Another person should be able to identify the intended measurement without guessing which stop, container, or unit was meant.

Methods should be precise about decisions without pretending that every project has universal settings. A water-only exercise can specify clean water, a pipette range, a target volume, tip type, aspiration and dispense actions, and a record template. A method for an unknown or hazardous material needs reviewed compatibility, risk assessment, waste, supervision, and equipment-specific instructions that this introductory lesson does not supply.

Write what changed

A deviation belongs beside the step it affects and in the deviation record, not hidden in a polished rewrite. If a different pipette was used, record its ID and reason. If a delay occurred, record timing and sample status. This allows a reader to distinguish the planned protocol from the actual run and to decide whether a repeat is needed. NIH reporting guidance emphasizes enough detail to distinguish technical from biological replicates and to make methods and data available for scrutiny.

Worked example

From slogan to method

Vague: ‘Pipette 1 mL water carefully.’ Revised: ‘Using P-02 and a compatible tip, set 1.00 mL. Pre-wet with clean water one to three times per the manual. Aspirate at the first stop with the tip vertical, dispense against the tube wall to the second stop, and record bubbles or droplets.’

Method

Work through the steps.

    State purpose and scope

    Name the measurand, material, and what the method can and cannot support.

  2. 02

    List identity and equipment

    Record sample or water identity, vessel, instrument type and ID, consumable type, and method or manual version.

  3. 03

    Number observable actions

    Write actions in order with units, timing, conditions, and endpoint observations. Avoid adjectives such as ‘carefully’ without a behavior.

  4. 04

    Add quality decisions

    State blanks, references, checks, acceptance criteria, and the action for a failure before the run.

  5. 05

    Link the record

    Provide a raw-data location, calculation trail, deviations, and revision date so the method and run can be connected.

Common mistakes

Correct the process, preserve the record.

‘Room temperature’ is used with no context.

Record the observed or specified range and whether temperature was controlled or merely noted.

A brand or model is listed without the relevant manual version.

Include the model, ID, source manual, and the exact instruction used where it affects operation.

Replicates are described as ‘n = 3.’

Say whether they are independent units or repeated readings of one unit and how they were ordered.

A deviation is edited out of the method.

Keep the approved plan and document the actual departure separately.

Rewrite a vague procedure

Turn ‘transfer water carefully and record the result’ into a numbered method that another beginner could follow safely with a permitted pipette.
